目的:观察通络止痛合剂对痛风性关节炎大鼠基质金属蛋白酶-3(MMP-3)、γ-干扰素(IFN-γ)的影响。方法:应用Coderre经典造模方法造模,予通络止痛合剂治疗,并以秋水仙碱、醋氯芬酸为对照,观察大鼠的一般状态、受试关节步态、关节肿胀周径及组织病理学的变化;采用ELISA法,检测各组大鼠受试关节内MMP-3、IFN-γ的含量;同时观察各治疗组对小鼠扭体实验的镇痛作用。结果:各治疗组均能明显改善关节滑膜炎症的病理改变、降低关节内MMP-3及IFN-γ的含量和减轻醋酸引起的小鼠疼痛(P<0.05);但各治疗组之间相比无差异(P>0.05)。结论:通络止痛合剂具有良好的消肿止痛作用、并通过抑制关节内MMP-3及IFN-γ的活性,减轻关节组织的炎症反应,以治疗痛风性关节炎。
Objective: To observe the effect of Tongluzhitong Mixture on the expression of matrix metalloproteinase-3 (MMP-3) and interferon-γ (IFN-γ) in gouty arthritis rats. Methods: The classic model of Coderre was used to make the model. The rats were treated with Tongluo Zhitong Mixture. Colchicine and aceclofenac were used as control. The general state of the rats, the gait of the joint under test, The pathological changes were observed. The contents of MMP-3 and IFN-γ in the joints of the rats in each group were detected by ELISA. The analgesic effects of each treatment group on writhing in mice were also observed. Results: Each treatment group could significantly improve the pathological changes of synovial inflammation, reduce the content of MMP-3 and IFN-γ in the joints and relieve the pain induced by acetic acid in mice (P <0.05). However, No difference (P> 0.05). Conclusion: Tonglu Zhitong Mixture has a good swelling and analgesic effect, and can reduce the inflammation reaction of joint tissues by inhibiting the activity of MMP-3 and IFN-γ in the joints and treating gouty arthritis.
