目的建立高效液相色谱同时测定“改善睡眠颗粒”中天麻素、红景天苷、栀子苷和斯皮诺素含量的方法。方法检测对象为以上提到的“改善睡眠颗粒”中的四个主要成分。采用的色谱柱为Thermo Fisher Syncronis C18(250 mm×4.6 mm,5μm);流动相为乙腈-水,梯度洗脱,流速为1.0 ml/min;柱温为30℃,进样量为10μl。检测波长分别设定为220、238、275、335 nm。结果天麻素、红景天苷、栀子苷和斯皮诺素分别在0.8032~4.0160、0.5392~2.6960、2.6916~13.4580、0.2112~1.0560μg范围内线性关系良好;精密度、稳定性、重复性试验的均RSD<2.00%;加样回收率分别为98.17%、99.37%、98.84%、99.27%;RSD分别为0.67%、0.48%、0.49%、0.93%。结论高效液相色谱法可同时测定“改善睡眠颗粒”中天麻素、红景天苷、栀子苷和斯皮诺素的含量,且该方法简便、准确,可用于该制剂的质量控制。
OBJECTIVE To establish a method for the simultaneous determination of gastrodin, salidroside, geniposide and spinosin in “Improving sleep particles” by high performance liquid chromatography. Methods The subjects were tested for the four major components in the above mentioned “Improving sleep particles.” The column used was Thermo Fisher Scientific Syncronis C18 (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of acetonitrile-water with gradient elution at a flow rate of 1.0 ml / min. The column temperature was 30 ℃ and the injection volume was 10μl. Detection wavelength was set to 220,238,275,335 nm. Results Gastrodin, salidroside, geniposide and spinosin showed good linearity in the range of 0.8032 ~ 4.0160,0.5392 ~ 2.6960,2.6916 ~ 13.4580,0.2112 ~ 1.0560μg, respectively. The precision, stability and reproducibility RSD <2.00%. The average recoveries were 98.17%, 99.37%, 98.84% and 99.27%, respectively. The RSDs were 0.67%, 0.48%, 0.49% and 0.93%, respectively. Conclusion The HPLC method can simultaneously determine the content of gastrodin, salidroside, geniposide and spinosin in “improving sleep granules”, and the method is simple and accurate and can be used for the quality control of the preparation .
