目的探讨TPX2蛋白和mRNA在宫颈鳞癌组织中的表达及其与宫颈鳞癌患者临床病理资料之间的相关性。方法 2009年10月至2010年11月在河北医科大学第四医院,采用免疫组织化学法、逆转录聚合酶链式反应(RT-PCR)法,对62例宫颈癌、15例正常宫颈组织和30例宫颈上皮内瘤变(CIN)组织进行检测,分析TPX2的mRNA和蛋白质的表达。结果经RT-PCR检测,正常宫颈组织中几乎不表达TPX2mRNA,宫颈鳞癌组织中相对含量为0.441±0.011,两者之间差异具有统计学意义(t=-19.568,P<0.01)。经免疫组化检测,TPX2蛋白在正常宫颈鳞状上皮组织中不表达,在CIN和宫颈鳞癌组织中呈阳性表达,三者之间差异具有统计学意义(P<0.05)。宫颈鳞癌组织中TPX2的表达与组织学分级、FIGO分期及淋巴结转移密切相关(P<0.05),而与年龄无关(P>0.05)。结论 TPX2可能参与了宫颈鳞癌的发生和发展,TPX2可作为宫颈鳞癌早期诊断及预后估计的一个指标。
Objective To investigate the expression of TPX2 protein and mRNA in cervical squamous cell carcinoma and its relationship with clinicopathological data of patients with cervical squamous cell carcinoma. Methods From October 2009 to November 2010 at the Fourth Hospital of Hebei Medical University, immunohistochemistry and reverse transcription polymerase chain reaction (RT-PCR) were performed on 62 cases of cervical cancer, 15 cases of normal cervical tissue and 30 cases of cervical intraepithelial neoplasia (CIN) tissues were detected, TPX2 mRNA and protein expression. Results There was almost no expression of TPX2 mRNA in normal cervical tissue by RT-PCR. The relative content of TPX2 in cervical squamous cell carcinoma tissue was 0.441 ± 0.011. The difference was statistically significant (t = -19.568, P <0.01). Immunohistochemistry showed that TPX2 protein was not expressed in normal cervical squamous epithelium and positive in CIN and cervical squamous cell carcinoma tissues, with statistical significance (P <0.05). The expression of TPX2 in cervical squamous cell carcinoma was closely related to histological grade, FIGO stage and lymph node metastasis (P <0.05), but not to age (P> 0.05). Conclusion TPX2 may be involved in the occurrence and development of cervical squamous cell carcinoma. TPX2 may serve as an indicator of early diagnosis and prognosis of cervical squamous cell carcinoma.
