液相色谱分析氨基酸邻苯二醛衍生物的方法已有不少报导,大都采用反相色谱柱前衍生法和pH梯度、离子浓度梯度并行的阳离子交换法。反相色谱柱前衍生法的优点是灵敏度高、分析时间短,但其缺点是衍生剂邻苯二醛必须输送柱内,这就严重地污染了柱子,使柱寿命迅速下降。pH梯度和离子浓度梯度并行法的优点是某些碱性氨基酸的分辨率高,但由于离子浓度较高,会磨损高压泵系统的密封垫,体系平衡的时间也较长。
Many methods have been reported for the liquid chromatographic analysis of o-phthalaldehyde derivatives of amino acids, mostly using reversed-phase column pre-derivatization and cation exchange in parallel with a gradient of pH and concentration of ions. The advantages of the reversed-phase column pre-derivatization method are high sensitivity and short analysis time, but the disadvantage is that the phthalaldehyde derivative must be transported inside the column, which severely pollutes the column and causes the column lifetime to drop rapidly. The advantage of the gradient gradient and ion concentration gradient parallel approach is the high resolution of some basic amino acids, but because of the high ion concentration, the gasket of the high pressure pump system is worn out and the system equilibration time is longer.
