目的探讨虫干预骨髓间充质干细胞(BMSCs)成脂和成骨分化的作用机制。方法传3代BMSCs分为空白对照组、激素组、虫组,3组干预6 d后对碱性磷酸酶(ALP)、甘油三酯(TG)进行检测,用光镜和电镜观察3组BMSCs细胞的超微结构。结果 虫含药血清干预BM-SCs后,虫组甘油三酯含量较激素组明显降低,细胞ALP含量较激素组明显增加,激素可以促使BMSCs脂肪分化,抑制细胞代谢和分裂增殖能力。结论 虫含药血清可以抑制BMSCs成脂分化,促使BMSCs成骨分化。
Objective To investigate the mechanism of the intervention of pinworm on adipogenic and osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs). Methods The third generation BMSCs were divided into blank control group, hormone group and pinworm group. The levels of alkaline phosphatase (ALP) and triglyceride (TG) in the three groups were detected after 6 d of intervention. Three groups were observed by light microscope and electron microscope Ultrastructure of BMSCs. Results The results showed that the triglyceride content in the pinworm group was significantly lower than that in the hormone group and the ALP content in the pinworm group was significantly higher than that in the hormone group after the intervention of the pinworm drug-containing serum. The hormone could promote the adipose differentiation of BMSCs and inhibit the cell metabolism and division and proliferation ability. Conclusions Serum containing pinworm can inhibit the adipogenic differentiation of BMSCs and promote the osteogenic differentiation of BMSCs.
