目的:研究通络救脑注射液及其2组分三七总皂苷、栀子苷对拟缺血损伤大鼠脑微血管内皮细胞脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)表达的影响。方法:采用原代培养大鼠脑微血管内皮细胞,氧糖剥夺法制备拟缺血损伤模型,三七总皂苷、栀子苷以及通络救脑注射液干预后,采用免疫蛋白印迹法和细胞免疫荧光法检测BDNF的表达情况。结果:与模型组比较,栀子苷对BDNF的上调作用不明显,三七总皂苷和通络救脑注射液可显著上调BDNF的表达,通络救脑注射液有优于2组分的作用趋势。结论:通络救脑注射液及其有效组分具有一定的促进BDNF表达的作用,这可能是该注射液在缺血性脑损伤后保护微血管和促进血管新生的内在机制之一。注射液的作用优于2组分,体现了中药复方的配伍优势。
Objective: To investigate the effects of Tongluo Jianyu injection and its two components, Panax notoginseng saponins and geniposide on the expression of brain-derived neurotrophic factor (BDNF) in rat brain microvascular endothelial cells influences. Methods: The model of ischemic injury induced by primary cultured rat brain microvascular endothelial cells was prepared by oxygen-glucose deprivation. After the intervention of Panax notoginseng saponins, geniposide and Tongluo Jiuna injection, the immunoblotting and cellular immunity Fluorescence detection of BDNF expression. Results: Compared with model group, geniposide had no obvious up-regulation effect on BDNF. Panax notoginseng saponin and Tongluojiannao injection could significantly up-regulate the expression of BDNF. Tonglianjuenao injection was superior to 2-component trend. Conclusion: Tongluo Jiuna injection and its effective components may promote the expression of BDNF, which may be one of the intrinsic mechanisms of the injection in protecting microvessels and promoting angiogenesis after ischemic brain injury. The role of injection is better than 2 components, reflecting the compatibility advantages of traditional Chinese medicine compound.
