目的建立大鼠子宫内膜异位症模型,探讨来氟米特抑制子宫内膜异位病灶生长的作用机制,为从免疫方面治疗子宫内膜异位症提供依据。方法采用自体移植法建立大鼠子宫内膜异位症模型。建模3周后治疗组来氟米特35mg/(kg.d),对照组1ml/(kg.d)生理盐水,连用3周后测量用药前后病灶体积,用免疫组化SP检测方法,检测移植灶中VEGF及TNF-α的表达。结果治疗后治疗组的病灶体积明显小于对照组(P<0.05);治疗后VEGF及TNF-α在治疗组的表达明显低于对照组(P<0.05)。结论本实验显示来氟米特对大鼠异位子宫内膜的生长具有很强的抑制作用。为治疗子宫内膜异位症提供新的应用前景。
Objective To establish a rat model of endometriosis and investigate the mechanism of leflunomide inhibiting the growth of endometriosis lesions and provide basis for the immunotherapy of endometriosis. Methods Autologous transplantation was used to establish a rat endometriosis model. After 3 weeks of treatment, leflunomide 35mg / (kg · d) and control group 1ml / (kg · d) saline were given respectively. The volume of lesion before and after 3 weeks of treatment was measured by immunohistochemical SP method The expression of VEGF and TNF-α in the transplanted tumor. Results After treatment, the lesion size of the treatment group was significantly smaller than that of the control group (P <0.05). After treatment, the expression of VEGF and TNF-α in the treatment group was significantly lower than that in the control group (P <0.05). Conclusion This experiment shows that leflunomide has a strong inhibitory effect on the growth of ectopic endometrium in rats. It provides a new application prospect for the treatment of endometriosis.
