目的:探讨决明子对高同型半胱氨酸血症(Hhcy)兔粥样斑块凋亡基因表达的影响。方法:将50只家兔标准饲料适应性喂养1周后,随机分成对照组、模型组、决明子1组、决明子2组、立普妥组。采用高L-蛋氨酸建立兔Hhey血症模型,比较各组兔Hcy含量、免疫组化观察Bax、Bcl-2在斑块中表达的干预效果。结果:决明子组、立普妥组血浆Hcy含量上升幅度明显低于同时段模型组,决明子2组治疗效果优于1组。免疫组化显示,模型组Bax表达与正常组有显著差异(P<0.01),决明子组和立普妥组与模型组比较,血管壁平滑肌细胞、泡沫细胞的Bcl-2的表达上调,Bax的阳性表达率下调,与模型组比较有显著差异(P<0.01)。同时决明子组Bax/Bcl-2值降低。结论:决明子可通过下调Bax/Bcl-2表达干预Hhcy兔动脉粥样硬化斑块的形成,剂量大小有差别。
Objective: To investigate the effect of Cassia seed on the gene expression of apoptosis in high homocysteinemia (Hhcy) rabbits. Methods: Fifty rabbits were fed with standard diet adaptively for one week and then randomly divided into control group, model group, Cassia seed 1 group, Cassia 2 group and Lipitor group. High L-methionine was used to establish rabbit model of Hheyremia. Hcy content in each group was compared. Immunohistochemistry was used to observe the effect of Bax and Bcl-2 expression in plaque. Results: The levels of plasma Hcy in Cassia group and Lipitor group were significantly lower than those in the model group at the same time. The therapeutic effect of Cassia group 2 was better than that of the 1 group. Immunohistochemistry showed that Bax expression in model group was significantly different from that in normal group (P <0.01). Compared with model group, the expression of Bcl-2 in vascular smooth muscle cells and foam cells in Cassia group and Lipitor group were up-regulated The positive expression rate was lower than that in the model group (P <0.01). At the same time, Cassia Bax / Bcl-2 decreased. CONCLUSION: Cassia seed can intervene the formation of atherosclerotic plaque in Hhcy rabbits by down-regulating the expression of Bax / Bcl-2, with different dose sizes.
