精原干细胞(SSC)是雄性动物体内生精过程中起重要作用的精原细胞类型,它不仅具有干细胞特性,而且还能分化成精子,是雄性动物体内唯一能将遗传信息传递给子代的成体干细胞。SSC可在无外源基因操作下诱导生成胚胎样多潜能干细胞,再转化为其他类型细胞。因此,SSC被认为是合乎道德、伦理、安全、理想的多潜能干细胞。除此之外,SSC的体外长期培养体系的建立、鉴定为移植和多潜能的研究提供了基础。早期SSC的研究主要在非灵长类动物体内进行。近几年,灵长类动物SSC的研究也取得了很大的进展,尤其是对人SSC的研究。本文主要对灵长类动物SSC的生物学特性、分离、纯化、培养、鉴定以及生物学标记进行简要的概述,并且对其无限多潜能性及其应用进行了讨论,进一步提出了SSC在男性不育症治疗和人类再生医学领域的应用潜能展望。
Spermatogonial stem cells (SSCs) are the types of spermatogonia that play an important role in the spermatogenesis of male animals. They not only have the characteristics of stem cells, but also differentiate into spermatozoa, which is the only way for male animals to transmit genetic information to offspring Adult stem cells. SSC induces the generation of embryonic pluripotent stem cells in the absence of exogenous gene manipulation and transforms into other types of cells. Therefore, SSC is considered to be an ethical, ethical, safe and ideal pluripotent stem cell. In addition, the establishment and identification of SSC long-term culture system in vitro provided the basis for the study of transplantation and multipotency. Early SSC research is mainly done in non-primate animals. In recent years, great progress has been made in the study of primate SSC, especially in the study of human SSC. In this paper, a brief overview of the biological characteristics, isolation, purification, culture, identification and biological markers of primate SSC has been given, as well as its unlimited potential and its applications were discussed. It is further proposed that SSC in male Fertility treatment and human regenerative medicine in the field of potential applications.
