目的观察慢性心理应激对2型糖尿病(T2DM)小鼠糖脂代谢及胰岛素抵抗(IR)的影响及加减逍遥散的作用效果。方法 102只自发性糖尿病(KK)小鼠随机分为5组:正常对照(WC)组,模型(SC)组,加减逍遥散对照(XYS)组,应激+加减逍遥散低剂量(LXYS)组及应激+加减逍遥散高剂量(HXYS)组。KK小鼠用高糖高脂饲料喂养8周,采用电击、夹尾、禁食、禁水、合笼饲养等方法复制慢性心理应激模型。检测各组小鼠空腹血糖(FPG)、血清胰岛素(FINS)以及血脂的胆固醇(CHO)、甘油三酯(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)4项等指标,计算胰岛素敏感指数(ISI)。结果与WC组比较,SC组的ISI明显降低(P<0.01),体质量明显下降(P<0.01),XYS组的FPG明显降低(P<0.01)、ISI明显增强(P<0.05);与SC组比较,LXYS组及HXYS组的ISI明显增强(P<0.01),HXYS组体质量明显升高(P<0.05),TG,CHO,LDL-C,明显降低(P<0.01,P<0.05),HDL-C,明显升高(P<0.05)。结论慢性心理应激能降低T2DM小鼠的胰岛素敏感性,从而加剧其IR以及糖脂代谢的紊乱;加减逍遥散不仅能提高T2DM小鼠的胰岛素敏感性,减轻IR,同时也能对抗慢性心理应激对T2DM小鼠IR的不良影响。
Objective To observe the effect of chronic psychological stress on the level of glucose and lipid metabolism and insulin resistance (IR) in type 2 diabetic mice and the effect of Xiaoyao San. Methods Totally 102 spontaneously diabetic (KK) mice were randomly divided into 5 groups: normal control (WC) group, model group (SC), XYS group, stress plus Xiaoyao Powder LXYS group and stress plus Xiaoyao powder high dose (HXYS) group. KK mice were fed with high-fat and high-fat diet for 8 weeks, and chronic psychological stress model was replicated by electric shock, tail-clamping, fasting, water forbidding and cage feeding. The levels of fasting blood glucose (FPG), serum insulin (FINS) and lipids in serum of the mice in each group were measured for cholesterol, triglyceride (TG), high density lipoprotein (HDL-C), low density lipoprotein (LDL- 4 items and other indicators, calculate the insulin sensitivity index (ISI). Results Compared with WC group, the ISI in SC group was significantly lower (P <0.01) and the body weight was significantly decreased (P <0.01). The FPG in XYS group was significantly lower (P <0.01) and ISI was significantly increased The ISI of LXYS group and HXYS group were significantly increased (P <0.01), the body weight of HXYS group was significantly increased (P <0.05), while TG, CHO and LDL-C were significantly decreased ), HDL-C, was significantly higher (P <0.05). Conclusion Chronic psychological stress can reduce the insulin sensitivity of T2DM mice, exacerbating the IR and glucose and lipid metabolism disorders; Xiaoyaosan can not only improve insulin sensitivity in T2DM mice, reduce IR, but also against chronic psychological Adverse Effects of Stress on IR in T2DM Mice.
