目的探讨血清脂联素水平及脂联素基因单核苷酸多态性(SNP)45T→G和276G→T两个位点与代谢综合征(MS)的关系。方法采用放免法对195例老年代谢综合征组(MS组)患者和156例对照组(Con组)人群进行血清脂联素水平测定,并用TaqMan探针技术进行脂联素基因SNP45分析,直接测序法进行脂联素基因SNP276分析。结果 MS组的血清脂联素水平较Con组显著降低(P<0.05);多元逐步回归分析显示:空腹胰岛素是影响MS患者血清脂联素水平的独立相关因素。脂联素基因多态性分析:SNP45在MS组与Con组比较差异有显著性(P<0.05);等位基因分布频率提示G等位基因在代谢综合征人群多见(P<0.05):MS组中携带TG+GG型的患者血清脂联素水平显著低于TT型的患者(P<0.05):Con组、MS组二组间SNP276基因型分布差异无统计学意义(P>0.05),等位基因分布频率分析显示无差异性(P>0.05)。结论低脂联素血症与MS的发生发展有关,SNP45 G/G基因型可能是中国汉族人MS的易感基因。
Objective To investigate the relationship between serum adiponectin level and single nucleotide polymorphisms (SNP) 45T → G and 276G → T of adiponectin and metabolic syndrome (MS). Methods Serum levels of adiponectin in 195 elderly patients with metabolic syndrome (MS group) and 156 healthy controls (Con group) were determined by radioimmunoassay. Adiponectin gene SNP45 was analyzed by TaqMan probe technique and sequenced directly Method of adiponectin gene SNP276 analysis. Results The level of serum adiponectin in MS group was significantly lower than that in Con group (P <0.05). Multivariate stepwise regression analysis showed that fasting insulin was an independent factor influencing serum adiponectin level in MS patients. Adiponectin gene polymorphism analysis showed that SNP45 was significantly different between MS group and Con group (P <0.05). Allele distribution frequency suggested that G allele was more common in the metabolic syndrome population (P <0.05) The levels of serum adiponectin in TG + GG patients in MS group were significantly lower than those in TT patients (P <0.05). There was no significant difference in SNP276 genotype distribution between Con group and MS group (P> 0.05) Allele distribution frequency analysis showed no difference (P> 0.05). Conclusion Low adiponectin is associated with the occurrence and development of MS. The SNP45 G / G genotype may be the susceptibility gene of MS in Han Chinese.
