通过茎段离体培养建立了四倍体刺槐无性系的微体繁殖体系。结果表明 :基本培养基为MS或WPM培养基。BA、NAA影响芽的增殖生长 ,在一定的范围内 ,BA对芽的增殖影响比NAA大 ,而NAA对芽高的影响比BA大 ,二者的比例对芽的增殖生长也有影响 ,芽增殖生长的最适BA和NAA组合应为BA0 5mg/L +NAA0 1mg L。四倍体刺槐无性系生根的最适生长调节物质配比为NAA0 2 5mg L +IBA0 4mg L。通过植株叶解剖观察 ,进一步证明了分步练苗能提高移栽成活率。
Micropropagation system of tetraploid black locust clones was established by in vitro culture of stem segments. The results showed that the basic medium was MS or WPM medium. BA and NAA affected bud proliferation and growth. Within a certain range, BA had a greater effect on bud proliferation than NAA, while NAA had a greater effect on shoot height than BA. The ratio of BA and NAA also had effects on the bud proliferation and proliferation. The optimal BA and NAA combinations for growth should be BA0 5 mg / L + NAA0 1 mg L. The optimal growth-regulating substance ratio for rooting of tetraploid black locust clones was NAA0 25mg L + IBA0 4mg L. By plant leaf anatomical observation, further evidence that step-by-step seedling can improve the survival rate of transplanting.
