探讨枇杷叶中熊果酸(UA)对人肝癌HepG2、人胃癌MGC803、BGC823以及人乳腺癌M231细胞致凋亡作用及其对凋亡相关的半胱天冬酶-3活性(Caspase-3)的影响。方法:不同浓度(10、20、40、50、80μmol/L)的UA作用细胞,采用噻唑蓝(MTT)比色法检测细胞增殖,普通光镜和AO/EB荧光染色法观察细胞凋亡形态学变化,琼脂糖凝胶电泳检测细胞凋亡梯度,比色法检测细胞凋亡过程中Caspase-3活性变化。结果显示不同浓度的UA组细胞的生长与对照组相比均受到不同程度的抑制,细胞增殖抑制率具有剂量依赖关系;普通光镜观察显示,给药组部分细胞呈典型的凋亡表现;AO/EB荧光染色可见核浓缩及核碎裂等典型的细胞凋亡特征;琼脂糖凝胶电泳可见典型的梯状条带;不同浓度UA组细胞中Caspase-3酶活性较对照组明显升高(P<0.05),其作用具有剂量依赖性。结论:UA可诱导上述四种癌细胞的生长,诱导细胞凋亡发生,Caspase-3的激活在UA诱导细胞凋亡机制中起到重要作用。
To investigate the effect of ursolic acid (UA) on the apoptosis of human hepatocellular carcinoma HepG2, human gastric cancer MGC803, BGC823 and human breast cancer M231 cells and its influence on apoptosis-related caspase-3 . Methods: UA-treated cells at various concentrations (10, 20, 40, 50 and 80 μmol / L) were cultured in vitro. Cell proliferation was detected by MTT assay. The changes of apoptosis were detected by agarose gel electrophoresis. The changes of Caspase-3 activity were detected by colorimetric assay. The results showed that the growth of UA group at different concentrations was inhibited to some extent compared with the control group, and the rate of cell proliferation inhibition was dose-dependent. The normal light microscopy showed that the apoptosis of some cells in the UA group was typical. / EB fluorescence staining showed typical characteristics of apoptosis such as nuclear condensation and nuclear fragmentation; typical ladder-shaped bands were observed by agarose gel electrophoresis; Caspase-3 activity in UA group was significantly higher than that in control group P <0.05), the effect of a dose-dependent manner. CONCLUSION: UA can induce the growth of the above four kinds of cancer cells and induce apoptosis. The activation of Caspase-3 plays an important role in the mechanism of UA-induced apoptosis.
