目的:探讨程氏分清饮对大鼠肾病综合征的作用机制。方法:采用大鼠尾静脉一次性注射盐酸多柔比星(5.5mg/kg)的方法造成大鼠肾病综合征模型,将实验动物随机分为正常对照组(A)、模型对照组(B)、程氏分清饮治疗组(C组)、激素治疗组(D组)、程氏分清饮加激素治疗组(E组)。观察程氏分清饮对肾病大鼠尿蛋白、血清白蛋白(Alb)、尿素氮(BUN)、血清肌酐(Cr)、甘油三酯(TG)、肾组织病理形态学的影响。结果:各治疗组24h尿蛋白明显低于B组(P<0.05,P<0.01);各治疗组血清生化指标较B组明显改善(P<0.05);肾组织病理改变明显,各治疗组MMP-9的表达明显高于B组(P<0.05)。结论:程氏分清饮通过上调MMP-9的表达,促进肾小球细胞外基质降解,防止肾小球硬化,同时可以降低肾病大鼠的尿蛋白,提高Alb、降低TG,阻止BUN、Cr的增高,修复足突融合,减轻肾组织损伤,保护肾功能。
Objective: To investigate the mechanism of Cheng’s Fenqingqing on rats with nephrotic syndrome. Methods: The rat model of nephrotic syndrome was induced by a single injection of doxorubicine hydrochloride (5.5mg / kg) into the tail vein of rats. The experimental animals were randomly divided into normal control group (A), model control group (B) , Cheng’s clear drink treatment group (C group), hormone treatment group (D group), Cheng’s clear drink plus hormone treatment group (E group). To observe the effect of Cheng’s Shixiangyin Decoction on urinary protein, serum albumin (Alb), blood urea nitrogen (BUN), serum creatinine (Cr), triglyceride (TG) and renal histopathology in rats with nephropathy. Results: The 24-hour urinary protein in each treatment group was significantly lower than that in group B (P <0.05, P <0.01). The serum biochemical indexes in each treatment group were significantly improved compared with those in group B (P <0.05) -9 expression was significantly higher than the B group (P <0.05). Conclusion: Cheng’s Decoction can up-regulate the expression of MMP-9, promote the glomerular extracellular matrix degradation, prevent glomerular sclerosis, and at the same time reduce the urinary protein in rats with nephrosis, increase Alb, lower TG, prevent BUN and Cr Increased, repair foot process fusion, reduce renal tissue damage, protect renal function.
