目的研究南海海绵Iotrochota sp.中的抗氧化活性成分。方法以95%乙醇浸提得南海海绵总浸膏,经多步分配得乙醇、正己烷和正丁醇部位。采用DPPH法进行活性示踪、反相柱层析和HPLC法分离,光谱方法进行结构鉴定。结果分离得到的化合物鉴定为purpurone,其清除DPPH自由基的IC50为19μg.m l-1。结论Purpurone为海绵Iotrochota sp.的特征性成分和主要抗氧化活性成分,对Iotrochota属海绵的分类具有重要意义。
Objective To study the antioxidant active ingredients of sponge Iotrochota sp. in the South China Sea. Methods The total extract of Nanhai Sponge was extracted with 95% ethanol, and the ethanol, n-hexane and n-butanol fractions were obtained after multiple steps. DPPH method was used to carry out active tracer, reversed phase column chromatography and HPLC separation, spectral identification of the structure. As a result, the isolated compound was identified as purpurone, and its IC50 for scavenging DPPH radicals was 19 μg·ml −1 . [Conclusion] Purpurone is the characteristic component and the main antioxidant activity component of Iotrochota sp. and it is of great significance to the classification of Iotrochota spongiformis.
