为探讨肝癌不同部分组织中AAD的表达和改变规律。方法将自身配对的肝癌、癌周和远癌组织中总RNA和AAD纯化,并分析了肝匀浆中AAD及不同分子形式AAD比活性,并与对应组织中GGT进行了比较。结果肝癌组织总RNA浓度,癌组织明显低于癌周和远癌组织(P<0.01);总AAD比活性(U/g)在癌组织明显高于远癌组织(P<0.05);肝癌组织中膜结合性AAD升高,但差别不明显(P>0.05),而可溶性AAD在肝癌组织中明显地高于癌周和远癌组织(P<0.05)。结论资料提示肝癌组织可表达大量AAD,且以可溶性AAD升高为主。
To investigate the expression and changes of AAD in different parts of liver cancer. Methods Total RNA and AAD were purified from self-matched hepatocellular carcinoma, pericancerous and distant cancer tissues, and the specific activities of AAD and AAD in different molecular forms in the liver homogenate were analyzed and compared with that in the corresponding tissues. Results The total RNA concentration in HCC tissues was significantly lower than that in cancerous and distant cancer tissues (P<0.01). The total AAD specific activity (U/g) was significantly higher in cancer tissues than in distal cancer tissues (P<0.05). The membrane-associated AAD in hepatocellular carcinoma was increased, but the difference was not significant (P>0.05). However, soluble AAD was significantly higher in hepatocellular carcinoma than in the pericancerous and distant carcinoma tissues (P<0.05). Conclusions The data suggest that hepatocellular carcinoma can express a large amount of AAD, with the increase of soluble AAD.
