基于黄曲霉毒素B_1(AFB_1)对乙酰胆碱酯酶(AChE)的抑制作用,建立了抑制型酶传感器快速检测AFB_1的方法。对实验参数进行优化,得到PBS缓冲溶液的最优pH为7.3、戊二醛的最佳质量分数为0.5%、最佳抑制时间为18 min。在最优条件下,建立标准曲线,并对样品进行检测。该方法对AFB_1的检测线性范围为3.00-14.00μg/mL,线性方程为y=2.509x+13.857(R~2=0.994),检出限为0.86μg/mL,花生油中AFB_1的加标回收率为87.49%-109.51%,玉米中AFB_1的加标回收率为89.00%-105.50%。
Based on the inhibition of AChE by aflatoxin B_1 (AFB_1), a method of rapid detection of AFB_1 by an inhibitory enzyme sensor was established. The optimum pH of the PBS buffer solution was 7.3, the best mass fraction of glutaraldehyde was 0.5%, and the best inhibition time was 18 min. Under the best conditions, a standard curve is established and samples are tested. The linear range of this method for AFB_1 was 3.00-14.00 μg / mL and the linear equation was y = 2.509x + 13.857 (R ~ 2 = 0.994). The detection limit was 0.86 μg / mL. The recoveries of AFB_1 Was 87.49% -109.51%. The recoveries of AFB_1 in corn were 89.00% -105.50%.
