目的:探讨去甲斑蝥素(NCTD)对人白血病细胞株K562细胞增殖的抑制作用。方法:用流式细胞仪测定NCTD对K562细胞体外培养过程中细胞毒性、细胞凋亡及细胞周期的影响。结果:NCTD对K562细胞有较强的增殖抑制作用;在作用24小时后达到凋亡高峰;24小时各浓度NCTD作用后,G_1期细胞百分数均减少,S期与G_2+M期细胞百分数均增加,呈现G_2+M期阻滞现象。结论:NCTD可诱导K562细胞凋亡,抑制瘤细胞分裂增殖,且有明显的时间和剂量效应。
Objective: To investigate the inhibitory effect of norcantharidin (NCTD) on the proliferation of human leukemia cell line K562. Methods: The effects of NCTD on cytotoxicity, apoptosis and cell cycle of K562 cells in vitro were determined by flow cytometry. RESULTS: NCTD had a strong inhibitory effect on K562 cells and reached the peak of apoptosis after 24 hours. After 24 hours of concentration of NCTD, the percentage of cells in G 1 phase decreased, and the percentage of cells in S phase and G 2 +M phase increased. , showing G_2+M phase retardation. Conclusion: NCTD can induce apoptosis of K562 cells and inhibit the proliferation and proliferation of tumor cells with obvious time and dose effects.
