目的:为了比较大鼠足趾皮下注射福尔马林(formalin)和腹腔注射脂多糖(LPS)诱导下丘脑室旁核(PVN)内c-Fos蛋白表达的差异。方法:本实验采用免疫组织化学ABC双重细胞染色法,观测formalin和LPS应激后下丘脑室旁核大细胞部(mPVN)和小细胞部(pPVN)内c-Fos蛋白和精氨酸加压素(AVP)的定位和分布情况。结果:(1)与空白对照组比较,LPS剂量依赖性地诱导mPVN和pPVN内c-Fos蛋白表达(P<0.05);(2)与空白对照组比较,formalin剂量依赖性地诱导pPVN内c-Fos蛋白表达(P<0.05),但mPVN内c-Fos蛋白无明显增加;(3)当两组模型大鼠pPVN内c-Fos蛋白表达无明显差异时,与formalin组比较,LPS组mPVN内c-Fos蛋白表达明显增多(P<0.05),两者mPVN内的AVP表达无明显差异。结论:结果提示formalin和LPS均可引起下丘脑PVN神经元产生特异性反应,但LPS诱发的c-Fos阳性神经元分布相对广泛,结果提示下丘脑神经元的激活与刺激方式、强度等相关,单一模式、单一指标来揭示和衡量其功能状态是不完善的。
OBJECTIVE: To compare the differences of c-Fos protein expression in hypothalamic paraventricular nucleus (PVN) between formalin and peritoneal injections of lipopolysaccharide (LPS) in subcutaneous intima of rats. Methods: Immunocytochemical ABC double staining was used to observe the effects of formalin and LPS on c-Fos protein and arginine in paraventricular nucleus (mPVN) and small cell (pPVN) of hypothalamus (AVP) positioning and distribution. (1) Compared with the blank control group, LPS induced c-Fos protein expression in mPVN and pPVN in a dose-dependent manner (P <0.05); (2) Compared with the blank control group, formalin induced a dose- (P <0.05), but there was no significant increase in c-Fos protein in mPVN. (3) When the expression of c-Fos protein in pPVN was not significantly different between the two groups, compared with the formalin group, mPVN The expression of c-Fos protein was significantly increased (P <0.05). There was no significant difference in the expression of AVP in mPVN between the two groups. CONCLUSIONS: Both formalin and LPS can induce hypothalamic PVN neurons to produce specific responses. However, the distribution of c-Fos positive neurons in LPS is relatively extensive. The results suggest that the activation of hypothalamic neurons is related to the stimulation mode and intensity, A single model, a single indicator to reveal and measure the functional status is not perfect.
