目的观察体外培养过程中胎儿软骨终板细胞Ⅱ型胶原和蛋白多糖变化的相关性。方法采用含体积分数10%胎牛血清的低糖DMEM培养基培养胎儿软骨终板细胞,应用Western blot和RT-PCR法检测第1代到第8代胎儿软骨终板细胞Ⅱ型胶原和蛋白多糖及其mRNA的表达情况,直线回归分析Ⅱ型胶原和蛋白多糖及其mRNA表达的相关性。结果第1~8代培养细胞的Ⅱ型胶原及蛋白多糖mRNA及蛋白表达水平两两比较差异均有统计学意义(P<0.05);Ⅱ型胶原及其mRNA表达与蛋白多糖及其mRNA的表达呈明显正相关(r=0.994 7,P=0.014;r=0.945 3,P=0.017)。结论抑制Ⅱ型胶原或蛋白多糖中一种物质下降可同时阻止另一种物质下降,从而延缓椎间盘退变发生。
Objective To observe the correlation between changes of type Ⅱ collagen and proteoglycan during fetal cartilage end-plate cells cultured in vitro. Methods Fetal cartilage end-plate cells were cultured in low glucose DMEM medium containing 10% fetal bovine serum. Western blot and RT-PCR were used to detect the type Ⅱ collagen and proteoglycan of fetal cartilage end-plate cells Its mRNA expression, linear regression analysis of type II collagen and proteoglycan and its mRNA expression correlation. Results The mRNA and protein expressions of type Ⅱ collagen and proteoglycan in the cultured cells of 1st to 8th generation were significantly different (P <0.05), the expressions of type Ⅱ collagen and mRNA and the expression of proteoglycan and mRNA There was a significant positive correlation (r = 0.994 7, P = 0.014; r = 0.945 3, P = 0.017). Conclusion Inhibition of type II collagen or proteoglycan in a substance decreased while preventing the decline of another substance, thus delaying the occurrence of disc degeneration.
