目的:检测醛固酮(Ald)诱导的心肌成纤维细胞(CFs)增殖中钙调神经磷酸酶(CaN)mRNA,蛋白表达及螺内酯(Spi),环孢素A(CsA)对其表达的影响,从而探讨CaN在Ald致心肌纤维化中的作用.方法:本研究以体外培养的新生SD大鼠CFs为研究对象,以Ald诱导其增殖和胶原合成,并加入CsA,Spi干预,应用RT-PCR和Western Blot分别测定CaNmRNA和蛋白表达的变化.结果:与对照组相比,Ald组促进CFs增殖和增加羟脯氨酸含量(P<0.05),与Ald组相比,Ald+CsA组,Ald+Spi组则可以明显抑制CFs增殖和减少羟脯氨酸含量(P<0.05);与对照组相比,Ald组CaNmR-NA和蛋白表达呈浓度依赖性升高(P<0.05),与Ald组比较,Ald+CsA组,Ald+Spi组的CaNmRNA,蛋白表达均降低(P<0.05).结论:外源性Ald可通过CaN依赖的信号转导通路诱导心肌纤维化,其作用与Ald的基因组通路有关.
AIM: To investigate the effects of aldosterone (Ald) on the expression of calcineurin (CaN) mRNA and protein, and the effect of spironolactone and CsA on the proliferation of cardiac fibroblasts (CFs) To explore the role of CaN in Ald induced myocardial fibrosis.Methods: CFs from SD rats were cultured in vitro and treated with Ald to induce proliferation and collagen synthesis. Western Blot were used to detect the changes of CaN mRNA and protein expression.Results: Compared with the control group, the Ald group promoted the proliferation of CFs and increased the hydroxyproline content (P <0.05). Compared with the Ald group, Ald + CsA group, Ald + Compared with the control group, the expression of CaNmR-NA and protein in Aldrich group was increased in a concentration-dependent manner (P <0.05), but not significantly different from that in Ald group (P <0.05). Spi group could significantly inhibit the proliferation of CFs and decrease the hydroxyproline content (P <0.05) .Conclusion: Exogenous Ald can induce myocardial fibrosis through CaN-dependent signal transduction pathway, and its effect is similar to Ald’s genomic Path related.
