目的探讨血管内皮生长因子(VEGF)和基质金属蛋白酶-9(MMP-9)与子痫前期发病的相关性。方法纳入江苏省常州市武进人民医院2013年1月-2016年11月确诊为子痫前期的患者100例,其中重度子痫前期患者49例,轻度子痫前期的患者51例,同时纳入同期分娩的健康孕妇50例作为对照组。采用Western blotting、半定量逆转录聚合酶链反应(RTPCR)分子生物学方法检测3组患者胎盘组织中VEGF、MMP-9的表达水平;采用酶联免疫吸附法(ELISA)检查患者血清及尿液中VEGF、MMP-9水平。结果 Western blotting、RT-PCR方法检测结果:轻、重度子痫前期组患者胎盘组织的VEGF、MMP-9表达水平均低于正常对照组,组间相比差异具有统计学意义(P<0.05);ELISA检测结果:两组血清及尿液中VEGF、MMP-9水平均低于正常对照组,且组间相比差异具有统计学意义(P<0.05)。结论 VEGF、MMP-9参与了子痫前期的病理生理改变,且与病情的发生及轻(重)程度有密切的相关性。
Objective To investigate the correlation between the expression of vascular endothelial growth factor (VEGF) and matrix metalloproteinase-9 (MMP-9) in preeclampsia. Methods One hundred patients diagnosed as preeclampsia from Wujin People’s Hospital of Changzhou, Jiangsu Province from January 2013 to November 2016 were enrolled. Among them, 49 cases were severe preeclampsia and 51 cases were mild preeclampsia, Fifty healthy pregnant women delivered as a control group. Western blotting and semi-quantitative reverse transcriptase-polymerase chain reaction (RTPCR) were used to detect the expression of VEGF and MMP-9 in the placenta of the three groups. Serum and urine were detected by enzyme-linked immunosorbent assay (ELISA) VEGF, MMP-9 levels. Results The results of Western blotting and RT-PCR showed that the expression levels of VEGF and MMP-9 in placenta of patients with mild and severe preeclampsia were significantly lower than those of normal control group (P <0.05) The results of ELISA showed that the levels of VEGF and MMP-9 in serum and urine in both groups were lower than those in normal control group (P <0.05). Conclusions VEGF and MMP-9 are involved in the pathophysiological changes of preeclampsia and are closely related to the occurrence and severity of the disease.
