目的 :研究复方中药对 Burkitt′s淋巴瘤细胞株 Raji细胞的影响 ,并探讨其机制。方法 :采用 MTT法检测细胞毒作用 ,利用 Wright- Gimesa染色法 ,Hoechst 33342荧光染色等手段观察凋亡细胞的形态结构变化 ,应用透射电镜技术和流式细胞分光光度术进行凋亡定性定量观察 ,并探讨凋亡抑制基因 bcl- 2蛋白表达。结果 :复方中药对 Raji细胞具有明显的生长抑制作用。共同孵育一段时间后 ,细胞呈现典型的凋亡形态学变化。流式细胞仪分析表明复方中药能使 Raji细胞发生凋亡 ,凋亡率呈现对浓度和时间的依赖性 ,并发现复方中药使 Raji细胞 bcl- 2基因的表达下调。结论 :复方中药能诱导 Raji细胞发生凋亡 ,其作用机制与 bcl- 2基因表达的下调有关。
Objective: To study the effect of compound Chinese herbs on Raji cells of Burkitt’s lymphoma cell line and to explore its mechanism. Methods: The cytotoxicity was detected by MTT assay. The morphological changes of apoptotic cells were observed by Wright-Gimesa staining and Hoechst 33342 fluorescence staining. The apoptosis and apoptosis were observed by transmission electron microscopy and flow cytometry. And to explore the expression of apoptosis-inhibiting gene bcl-2 protein. Results: Compound Chinese herbs had obvious growth inhibitory effect on Raji cells. After co-incubation for a period of time, the cells showed typical apoptotic morphological changes. Flow cytometry analysis showed that compound Chinese herbs could induce apoptosis of Raji cells, and the apoptosis rate showed a concentration-dependent and time-dependent manner. It was found that the compound Chinese herbs down-regulated the expression of bcl-2 gene in Raji cells. Conclusion: Compound Chinese herbal medicine can induce apoptosis in Raji cells. Its mechanism is related to the down-regulation of bcl-2 gene expression.
