目的:了解加味桂枝茯苓颗粒对大鼠良性前列腺增生症微血管密度(MVD)计数、bcl-2基因表达的影响,以期明确加味桂枝茯苓颗粒对前列腺新生血管产生和基质细胞生长干预的分子生物学机制。方法:大鼠80只,分为空白对照组,空白增生组,加味桂枝茯苓颗粒组和非那雄胺组。造模成功后,对应组分别采用不给药,0.9%氯化钠溶液,加味桂枝茯苓颗粒悬液,非那雄胺溶液灌胃治疗。后观察前列腺组织生长情况,免疫组化检测前列腺组织MVD,FCM检测bcl-2基因表达。结果:加味桂枝茯苓颗粒组和非那雄胺组较空白增生组MVD计数、bcl-2表达量为均显著下降(P<0.01),且两组之间无明显差异。结论:加味桂枝茯苓颗粒可明显降低大鼠前列腺MVD计数、bcl-2的基因表达,减少前列腺组织微血管产生,加速腺细胞的凋亡,明显减少基质细胞产生,可能是其治疗前列腺增生的重要原因。
Objective: To understand the effect of Jiawei Guizhi Fuling granule on microvessel density (MVD) count and bcl-2 gene expression in rats with benign prostatic hyperplasia, in order to clarify the effect of Jiawei Fuling Zhuling Capsule on molecular biology of prostatic neovascularization and stromal cell growth Learning mechanism. Methods: Eighty rats were divided into blank control group, blank hyperplasia group, Jiawei Guizhifuling granule group and finasteride group. After the success of modeling, the corresponding groups were administered with no 0.9% sodium chloride solution, flavored Guizhi Fuling granules suspension, finasteride solution gavage treatment. The growth of prostate tissue was observed, the expression of bcl-2 gene was detected by immunohistochemistry in prostate tissue MVD and FCM. Results: Compared with blank hyperplasia group, the MVD count and bcl-2 expression in the Modified Guizhi Fuling Granule Group and Finasteride Group were significantly decreased (P <0.01), and there was no significant difference between the two groups. Conclusion: Modified Guizhi Fuling Granule can significantly reduce the prostate MVD count, bcl-2 gene expression, reduce prostate microvessel production, accelerate glandular cell apoptosis, significantly reduce stromal cell production may be an important reason for the treatment of benign prostatic hyperplasia .
