目的探讨脐血单个核细胞移植(Mesnchymal Stem Cells,MSCs)对大鼠脑梗死后神经功能的影响。方法将90只实验大鼠随机分成MSCs组、模型组;制备脑梗死模型;免疫组化检测Nestin、Shank1表达;电镜观察神经元超微结构并对神经突触进行定量分析;实验大鼠神经功能检测。结果 MSCs组Shank1及Nestin蛋白阳性表达升高(P<0.01)。MSCs组神经元及突触结构正常,突触数量、界面曲率及突触后致密区增大,突触间隙变窄(P<0.01)。模型组神经元形态异常、突触溶解、胞质空化、线粒体肿胀、细胞器减少。神经功能评分,神经行为学检测,梗死组不对称分值升高,MSCs组不对称分值较明显下降(P<0.01)。结论脐血单个核细胞移植可促进大鼠脑梗死后神经功能恢复。
Objective To investigate the effects of cord blood mononuclear cell transplantation (MSCs) on the neurological function after cerebral infarction in rats. Methods Ninety experimental rats were randomly divided into MSCs group and model group. The cerebral infarction model was prepared. The expression of Nestin and Shank1 was detected by immunohistochemistry. The ultrastructure of neurons was observed by electron microscope and the synapses were quantitatively analyzed. Detection. Results The positive expression of Shank1 and Nestin in MSCs group was significantly increased (P <0.01). The neurons and synapses in MSCs group were normal, the number of synapses, the curvature of interface and the area of postsynaptic density increased, and the synaptic cleft became narrower (P <0.01). Neurons in the model group were abnormal in morphology, synaptic lysis, cytoplasmic cavitation, mitochondria swelling, organelles decreased. Neurological score, neurobehavioral test, asymmetric score of infarction group increased significantly, and asymmetry score of MSCs group decreased significantly (P <0.01). Conclusion Umbilical cord blood mononuclear cells transplantation can promote neurological recovery after cerebral infarction in rats.
