目的 观察体外培养的正常猪视网膜色素上皮 (RPE)细胞中游离钙。方法 培养猪RPE细胞后 ,用激光扫描共聚焦显微镜 (LSCM)观察细胞中游离钙的分布特点 ,并通过校正求得游离钙的绝对值。结果 猪RPE细胞中游离钙的分布特点是细胞核较细胞浆中浓度高 ,通过校正求得细胞中游离钙平均值为 ( 2 2 0 .3 7± 2 7.3 5 )nmol/L。结论 用LSCM能直接观察培养的RPE细胞内游离钙的分布 ,并可求得胞内游离钙的平均绝对值
Objective To observe the free calcium in normal pig retinal pigment epithelium (RPE) cells cultured in vitro. Methods After pig RPE cells were cultured, the distribution characteristics of free calcium in cells were observed by laser scanning confocal microscope (LSCM), and the absolute value of free calcium was obtained by calibration. Results The distribution of free calcium in porcine RPE cells was characterized by high concentration in the cytoplasm of cytoplasm. The average value of the free calcium in the cells was (20.37 ± 2.73 5) nmol / L. Conclusion LSCM can directly observe the distribution of intracellular free calcium in cultured RPE cells and obtain the average absolute value of intracellular free calcium
