目的:建立测定细胞色素 C Y P2 D6 酶活性的毛细管气相色谱方法。方法:受试者口服右美沙芬( D M) 后收集0 ~8 h 尿样,经提取分离,以 H P1 毛细管柱分离, F I D 为检测器。结果:右美沙芬与代谢物3羟基 N甲基吗喃( D T) 分离良好, D M 在020 ~160 μg·m L- 1 , D T 在040 ~2000 μg·m L- 1 范围内线性关系良好,回收率分别为995 % 和998 % ,日内相对标准偏差分别小于547 % 和619 % , D M 和 D T 的最低检测限分别为8 ng 和4 ng 。用此法对11 名健康志愿者进行了 C Y P2 D6 活性测定。结论:此法简便、准确,可用于 C Y P2 D6 酶活性的测定。
Objective: To establish a capillary gas chromatographic method for the determination of cytochrome C Y P2 D6 enzyme activity. Methods: Subjects received dextromethorphan (D M) and collected urine samples for 0 to 8 hours. The samples were separated and separated by H P1 capillary column. F I D was the detector.
