目的探讨神经生长因子(NGF)β-疏基乙醇(BME)诱导小鼠骨髓间充质干细胞(mouse marrow mesenchymal stem cells, mMSCs)分化为神经元样细胞的作用。方法密度梯度离心结合差异贴壁法分离、纯化mMSCs,用血小板源性生长因子(Platelet Derived Growth Factor, PDGF-BB)和表皮生长因子(Epidermal Growth Facotro, EGF)刺激促进mMSCs分裂增殖。以NGF和BME为诱导剂,观察诱导期间细胞的形态变化,并用免疫细胞化学法鉴定神经元烯醇化酶(NSE)、神经丝蛋白(NF)、胶质纤维酸性蛋白(GFAP)的表达,甲苯胺蓝色法显示尼氏小体。结果免疫细胞化学显示:70%细胞表达NSE,74%细胞表达NF;实验组行尼氏染色出现尼氏小体。结论NGF和BME能诱导mMSCs 分化为神经元样细胞,而且具有较高的诱导效率。
Objective To investigate the effect of NGF β-mercaptoethanol (BME) on differentiation of mouse bone marrow mesenchymal stem cells (mMSCs) into neuron-like cells. Methods The mMSCs were isolated and purified by density gradient centrifugation and differential adherent method. The proliferation of mMSCs was stimulated by Platelet Derived Growth Factor (PDGF-BB) and Epidermal Growth Facotro (EGF). NGF and BME were used as inducers to observe the morphological changes of the cells during induction, and the expressions of neuron enolase (NSE), neurofilament (NF) and glial fibrillary acidic protein (GFAP) were identified by immunocytochemistry. Aniline blue shows Nissl bodies. Results Immunocytochemistry showed that NSE was expressed in 70% of cells and NF in 74% of cells. Nissl microscopy was performed in the experimental group. Conclusion NGF and BME can induce mMSCs to differentiate into neuron-like cells with high induction efficiency.
