目的探讨辛基酚(octylphenols,OP)诱导乳腺癌细胞凋亡及抑制乳腺癌细胞增殖的可能机制。方法应用MTT法及流式细胞仪检测OP对乳腺癌细胞MCF-7的增殖抑制及细胞周期的变化;Real Time PCR和Western blot法检测OP对MCF-7乳腺癌细胞中细胞周期相关蛋白p27Kip1和CyclinE的mRNA及蛋白表达的影响及对凋亡抑制蛋白PAK4表达的影响。结果 OP对MCF-7细胞增殖有明显的抑制作用,MCF-7细胞阻滞在G0/G1期,使S期的细胞减少;Real Time PCR和Western Blot检测发现培养的MCF-7细胞,随着OP的处理浓度增加,CyclinE蛋白在分子水平和蛋白水平均降低,p27Kip1的表达量升高。结论 OP可能通过调控细胞周期相关蛋白的表达,抑制MCF-7乳腺癌细胞增殖。
Objective To investigate the possible mechanism of octylphenols (OP) inducing apoptosis of breast cancer cells and inhibiting the proliferation of breast cancer cells. Methods MTT and flow cytometry were used to detect the effects of OP on the proliferation and cell cycle of MCF-7 breast cancer cells. Real-time PCR and Western blot were used to detect the expression of cell cycle-associated proteins p27Kip1 and p27Kip1 in MCF-7 breast cancer cells Effect of CyclinE mRNA and protein expression on the expression of PAK4 protein. Results The proliferation of MCF-7 cells was significantly inhibited by OP. MCF-7 cells were arrested in G0 / G1 phase and decreased in S phase. Real-time PCR and Western Blot showed that MCF-7 cells cultured with MCF- The concentration of OP increased, the level of CyclinE protein and protein decreased, and the expression of p27Kip1 increased. Conclusion OP may inhibit the proliferation of MCF-7 breast cancer cells by regulating the expression of cell cycle related proteins.
