目的:建立非水毛细管电泳法(NCAE)快速测定化痔栓中苦参碱的含量。方法:运用非水毛细管电泳方法,熔融石英毛细管(50μmID×50cm),缓冲液为70mmol.L-1醋酸铵-7.0%醋酸-10%乙腈的甲醇液,检测波长220nm,分离电压25kV,柱温20℃,用0.45μm微孔滤膜过滤后进样,压力进样:50mbar×3s。结果:方法最低检测为0.4μg·mL-1,线性范围5~500μg·mL-1,r=0.9991(n=7),线性关系良好;迁移时间的日内RSD为0.91%,日间RSD为1.52%;峰面积的日内RSD为1.67%,日间RSD为3.33%;平均回收率为99.04%。结论:为化痔栓中苦参碱的含量测定提供了一种快速、准确和灵敏的测定方法。
Objective: To establish a non-aqueous capillary electrophoresis (NCAE) method for rapid determination of matrine in Huatan suppository. Methods: Non-aqueous capillary electrophoresis method was used. Fused silica capillary (50μmID×50cm) was used. The buffer was 70mmol.L-1ammonium acetate-7.0%acetic acid-10% acetonitrile in methanol. The detection wavelength was 220nm. The separation voltage was 25kV. 20 °C, 0.45μm microporous membrane filter after injection, pressure injection: 50mbar × 3s. Results: The lowest detection method was 0.4μg·mL-1, the linear range was 5~500μg·mL-1, r=0.9991 (n=7). The linear relationship was good. The intraday RSD of the migration time was 0.91%, and the RSD was 1.52 during the day. %; The intraday RSD of the peak area was 1.67%, and the RSD was 3.33% during the day; the average recovery rate was 99.04%. Conclusion: A rapid, accurate and sensitive method for the determination of matrine in Huatan Suppository is provided.
