本文在用双垂直板等电聚焦-聚丙烯酰胺凝胶电泳(isoelectrofocusing-polyacrylamidegelelectrophoresis,IEF-PAGE)和双波长自动层析扫描仪对5个品种优质高蛋白稻米胚乳蛋白全组分作定性定量基础上,对其中的一个代表品种的胚乳全蛋白组分进行制备IEF-PAGE;经扫描定性定量后,按5个PI范围——PI8.6~7.8,PI7.4~6.6,PI6.6~5.8,PI5.8~5.0,PI5.0~4.0分别洗脱各蛋白组分;用氨基酸自动分析仪测定它们的氨基酸种类与含量;依FAO/WHO标准,计算各PI范围蛋白组分的必需氨基酸配比。结果表明,PI5.8~5.0蛋白组分的氨基酸配比最佳(营养价值最高),其次的是PI6.6~5.8,最差的是H7.4~6,6。本文与彻底弄清籼稻胚乳中最高营养的蛋白组分的氨基酸顺序,以便通过分子育种等途径更有效地改良稻米营养品质关系密切。
In this paper, the qualitative and quantitative basis of the whole components of five varieties of high quality and high protein rice endosperm protein were determined by dual-vertical plate isoelectric focusing-polyacrylamide gel electrophoresis (IEF-PAGE) and dual-wavelength automatic chromatographic scanner The whole endosperm protein component of one of the representative cultivars was prepared for IEF-PAGE. After scanning qualitative and quantitative analysis, five PI ranges of PI8.6 ~ 7.8, PI7.4 ~ 6.6, PI6 .6 ~ 5.8, PI5.8 ~ 5.0, PI5.0 ~ 4.0 were eluted from each protein components; amino acid analyzer with their amino acid species and content; according to FAO / WHO standard, calculated Essential amino acid ratio for each PI range protein component. The results showed that the amino acid composition of PI5.8 ~ 5.0 was the best (highest nutritional value), followed by PI6.6 ~ 5.8, and the worst was H7.4 ~ 6,6. In this paper, we thoroughly elucidate the amino acid sequence of the highest nutritional protein component in indica rice endosperm in order to improve the nutritional quality of rice more effectively through molecular breeding and other means.
