目的探讨金属硫蛋白(metallothionein,MT)对染铅小鼠的排铅作用。方法选择雄性昆明种小鼠40只,随机分为正常对照组,醋酸铅模型组,MT组(MT低、高剂量组),依地酸二钠钙组,共5组,每组8只。除正常对照组外,其余4组小鼠连续30d自由饮用0.55g/L醋酸铅水溶液制作铅中毒模型。MT低、高剂量组小鼠在染铅同时以不同剂量MT灌胃干预;依地酸二钠钙组小鼠染铅同时每天肌肉注射依地酸二钠钙。结果 MT组小鼠血铅和股骨铅含量明显低于醋酸铅模型组,差异有统计学意义(P<0.05);MT组血清中的丙二醛(MDA)含量低于醋酸铅模型组,而总抗氧化能力(T-AOC)含量高于醋酸铅模型组,差异有统计学意义(P<0.05)。结论金属硫蛋白具有明显的排铅作用和一定的抗氧化作用。
Objective To investigate the lead efflux of metallothionein (MT) on lead-exposed mice. Methods Forty male Kunming mice were randomly divided into normal control group, lead acetate model group, MT group (MT low and high dose group) and edetate disodium calcium group (n = 5), with 8 mice in each group. Except the normal control group, the other 4 groups of mice were allowed to drink 0.55 g / L lead acetate aqueous solution for 30 days to make lead poisoning model. MT low and high dose groups of mice in the lead at the same time with different doses of MT intragastric intervention; edetate disodium calcium group of mice lead by intramuscular injection of edetate calcium disodium. Results The levels of lead and lead in blood lead and femur of MT group were significantly lower than that of lead acetate group (P <0.05). The content of malondialdehyde (MDA) in serum of MT group was lower than that of lead acetate group Oxidation ability (T-AOC) content was higher than lead acetate model group, the difference was statistically significant (P <0.05). Conclusion Metallothionein has obvious lead excretion and certain anti-oxidative effects.
