目的:采用HPLC-DAD-ESI-MS分析芍药甘草配伍体内外物质基础变化,确认大鼠血中移行组分。方法:以Kor-masilC18色谱柱(4.6mm×250mm,5μm带保护柱);以乙腈-0.5%乙酸水为流动相进行梯度洗脱,流速0.8mL·min-1,柱温25℃,以甲醇为去蛋白溶剂,样品经电喷雾离子源正离子化后,通过LCQdeca型质谱仪,在m/z100~1000进行扫描,并对特征离子进行2次裂解,获得二级质谱数据。结果:以芍药甘草复方组方前后的体外成分研究为基础,确认了芍药甘草汤体外的主要成分为芍药苷、儿茶素-5-O-葡萄糖苷、白芍苷、甘草苷、甘草酸等,入血移行成分中也发现此类糖苷类物质,另又发现了来源于甘草成分的葡萄糖醛酸结合物——甘草素葡萄糖醛酸和异甘草酸葡萄糖醛酸。结论:采用液相色谱-质谱联用技术分析芍药甘草汤大鼠血中移行组分,并与芍药甘草复方组方前后的成分进行对比,可对复方入血的成分进行推测和确认,具有简便、快速的优点,且对于中药复方物质基础研究具有较好的应用价值。
OBJECTIVE: HPLC-DAD-ESI-MS was used to analyze the changes of the substance in vitro and in vivo of Peony licorice in vitro and in vivo. Conclusion: The composition of the blood in the rats of Shaoyao Gancao Decoction is analyzed by liquid chromatography-mass spectrometry, and compared with the ingredients before and after the compound prescription of Shaoyao Gancao, which can be used to estimate and confirm the composition of compound bleeds. , Rapid advantages, and for the basic research of traditional Chinese medicine compound has good application value.
