目的采用超高效液相色谱-电喷雾飞行时间质谱联用(UPLC-TOF/MS)测定苦荞中γ-氨基丁酸(GABA)的含量。方法 GABA与盐酸正丁醇衍生化反应形成γ-氨基丁酸丁酯的衍生物。色谱柱为BEH C18柱(100 mm×2.1 mm,1.7μm);流动相为乙腈-甲醇-0.01 mol·L-1乙酸铵溶液(45∶35∶30);流速0.2 mL·min-1;柱温30℃;采用飞行时间质谱仪电喷雾电离源(ESI)、正离子(V模式)方式检测。结果 GABA在0.4831~4.8312μg范围内制备的衍生物进样,平均加样回收率为97.28%(r=0.9996)。结论本法简便、准确、重现性好,可用于苦荞中GABA的含量测定。
Objective To determine the content of γ-aminobutyric acid (GABA) in buckwheat by ultra performance liquid chromatography-electrospray ionization time of flight mass spectrometry (UPLC-TOF / MS). Methods The derivatization of GABA with n-butanol hydrochloride to form butyl γ-aminobutyrate. The column was BEH C18 (100 mm × 2.1 mm, 1.7 μm). The mobile phase was acetonitrile-methanol-0.01 mol·L-1 ammonium acetate solution (45:35:30) and the flow rate was 0.2 mL · min-1. Temperature 30 ℃; using time of flight mass spectrometer electrospray ionization source (ESI), positive ion (V mode) detection. Results The derivatives of GABA prepared in the range of 0.4831 ~ 4.8312μg were injected with the average recovery of 97.28% (r = 0.9996). Conclusion The method is simple, accurate and reproducible. It can be used for the determination of GABA in tartary buckwheat.
