目的:研究乌头母根、子根、须根的多糖含量及单糖组成。方法:分别以精制多糖测得多糖对葡萄糖的换算因子,以苯酚-硫酸分光光度法测定乌头母根、子根、须根的多糖含量;以1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生高效液相色谱法测定多糖中的单糖组成。结果:乌头母根、子根、须根的多糖含量分别为22.0%,33.5%,6.1%;母根、子根、须根的多糖中主要含有葡萄糖,另外含有少量的半乳糖醛酸、半乳糖、阿拉伯糖,子根中还含有甘露糖,须根中还含有甘露糖、鼠李糖、木糖。结论:多糖含量检测方法简便、快速,测定结果更客观、准确;须根中多糖含量最低,且单糖组成与母根、子根有明显不同。
Objective: To study the content of polysaccharides and the composition of monosaccharide in root, root and root of aconite. Methods: The conversion factors of polysaccharides to glucose were measured with refined polysaccharides. The content of polysaccharides in root, root and fibrous roots of A. capitatum was determined by phenol-sulfuric acid spectrophotometry. Results: The contents of polysaccharides in root, root and fibrous roots of Acacia were 22.0%, 33.5% and 6.1%, respectively. The polysaccharides of root, root and root mainly contained glucose, and a small amount of galacturonic acid and galactose , Arabinose, sub-root also contains mannose, fibrous roots also contain mannose, rhamnose, xylose. Conclusion: The detection method of polysaccharide content is simple and rapid, the determination results are more objective and accurate. The content of polysaccharide in the fibrous roots is the lowest, and the composition of monosaccharides is obviously different from that of the parent roots and roots.
