目的 研究经三维培养后的主动脉平滑肌细胞 (ASMC) 胶原凝胶移植后 ,是否仍能产生弹性纤维及宿主的接受状态。 方法 将仿真皮替代物三维培养 2周的ASMC 胶原凝胶植入大鼠皮下 ,分别于术后 4、7、10、14和 2 8d取材 ,用HE染色、Gomori醛复红染色、α肌动蛋白免疫组织化学染色和弹性蛋白免疫组织化学染色 ,观察和检测ASMC 胶原凝胶。 结果 在移植后第 1周有大量的弹性纤维产生 ,但于 2 8d时完全消失。在移植后的 2 8d内 ,ASMC 胶原凝胶块周围没有明显的白细胞浸润。 结论 利用ASMC 胶原凝胶作为一种有弹性的真皮替代物 ,尚需进一步的研究。
OBJECTIVE: To study whether elastic fibers and host status can still be produced after three-dimensional culture of aorta smooth muscle cells (ASMC) collagen gel. Methods ASMC collagen gel three-dimensionally cultured for 2 weeks was implanted subcutaneously in rats. The rats were drawn at 4, 7, 10, 14 and 28 days after operation respectively. HE staining, Gomori aldehyde complex red staining, Protein immunohistochemical staining and elastin immunohistochemical staining were used to observe and detect ASMC collagen gel. Results In the first week after transplantation, a large amount of elastic fibers were produced, but disappeared completely after 28 days. Within 28 days after transplantation, there was no significant leukocyte infiltration around the ASMC collagen gel mass. Conclusion ASMC collagen gel as a flexible dermal substitute needs further study.
