目的:观察人参三七川芎提取物对老龄大鼠免疫器官功能及行为学的影响。方法:选取100只SD大鼠,饲养至18~19月龄,建立自然衰老大鼠模型。按体重水平分层后将成模大鼠随机分为5组,分别为衰老模型组、人参三七川芎中药低、中、高剂量组(2 305.64,1 152.82,576.41 mg·kg-1)和白藜芦醇组(50 mg·kg-1),另设2月龄SD大鼠做为青年对照组。干预3个月后,进行旷场实验,记录大鼠跨越方格总数和总行径距离,检测大鼠血清活性氧族(reactive oxygen species,ROS)含量,胸腺系数和脾脏系数。结果:与青年组大鼠比较,自然衰老组大鼠跨格总数减少(P<0.01),总行径距离减少(P<0.05),血清ROS含量增高(P<0.01),胸腺系数和脾脏指数下降(均P<0.01)。与衰老模型组比较,中药各组和白藜芦醇组大鼠的跨格次数和总行径距离均有增加(P<0.05);血清活性氧族(ROS)含量降低(P<0.05);中药高剂量组和白藜芦醇组可以增高大鼠的胸腺系数和脾脏系数(均P<0.01),中药中剂量组可以增高大鼠的胸腺系数(P<0.05)。结论:人参三七川芎提取物可以改善自然衰老大鼠精神迟缓状态,增强其空间探索性,同时能够提高免疫器官功能和降低血清活性氧含量。
Objective: To observe the effects of ginseng and rhizoma chuanxiong rhizome extract on immune function and behavior in aged rats. Methods: One hundred SD rats were selected and reared until 18-19 months old to establish a natural aging rat model. The rats were randomly divided into 5 groups according to their body weight: aging model group, low, medium and high dose group of Panax notoginseng and Chuanxiong (2 305.64,1 152.82,576.41 mg · kg-1) and white Resveratrol group (50 mg · kg-1), another two-month-old SD rats as a youth control group. After intervention for 3 months, the open-field test was carried out. The total number of squares and the total distance of the traverse path were recorded. The levels of reactive oxygen species (ROS), thymus and spleen in the rats were measured. Results: Compared with the young rats, the total number of cross-cells in the natural aging group decreased (P <0.01), the total distance traveled decreased (P <0.05), the serum ROS increased (P <0.01), the thymus index and the spleen index decreased (All P <0.01). Compared with aging model group, the number of cross-grids and total path length of rats in each group and resveratrol group increased (P <0.05), and the content of reactive oxygen species (ROS) decreased (P <0.05) The high dose group and resveratrol group could increase the thymus coefficient and spleen coefficient of rats (all P <0.01). The middle dose group of Chinese medicine can increase the thymus coefficient of rats (P <0.05). Conclusion: Ginseng Extract of Chuanxiong notoginseng can improve the state of mental retardation and enhance the space exploration of natural aging rats. At the same time, it can improve the function of immune organs and reduce the content of serum reactive oxygen species.
