目的检测ERG蛋白和鼠双微体(MDM2)蛋白在前列腺癌组织中的表达情况,并探讨其相关性。方法收集经病理学确诊为前列腺癌标本48例,良性前列腺增生10例,采用免疫组织化学二步法或SP法检测组织标本的ERG蛋白和MDM2蛋白的表达情况,采用χ~2检验及Spearman等级相关分析ERG和MDM2表达与前列腺癌分级的关系。结果前列腺癌组织中ERG和MDM2蛋白阳性表达率分别为33.3%和47.9%,在良性前列腺增生组未见表达,差异有统计学意义(P<0.05)。ERG和MDM2蛋白表达与前列腺癌Gleason评分分级不相关(P>0.05);但ERG与MDM2蛋白在前列腺癌组织中的表达呈正相关(r=0.831,P=0.001)。结论 ERG和MDM2蛋白在前列腺癌组织中的高表达,与前列腺癌的发生、发展密切相关。前列腺癌中ERG蛋白表达与MDM2蛋白的异常表达密切相关说明这两个蛋白可能存在协同作用。
Objective To detect the expression of ERG protein and murine diademosome (MDM2) protein in prostate cancer and to explore their correlation. Methods Forty-eight cases of prostate cancer and 10 cases of benign prostatic hyperplasia were collected by pathology. The expression of ERG protein and MDM2 protein were detected by immunohistochemical two-step method or SP method. The Chi-square test and Spearman grade Correlation Analysis ERG and MDM2 Expression and Prostate Cancer Classification. Results The positive expression rates of ERG and MDM2 in prostate cancer tissues were 33.3% and 47.9%, respectively, which were not found in benign prostatic hyperplasia group. The difference was statistically significant (P <0.05). The expression of ERG and MDM2 was not correlated with the Gleason score of prostate cancer (P> 0.05). However, there was a positive correlation between the expression of ERG and MDM2 in prostate cancer (r = 0.831, P = 0.001). Conclusion The high expression of ERG and MDM2 in prostate cancer tissues is closely related to the occurrence and development of prostate cancer. The close relationship between ERG protein expression and abnormal MDM2 protein expression in prostate cancer suggests that there may be synergistic effects between these two proteins.
