目的:研究滋阴抑亢调冲汤(zyyktc)对卵巢过度刺激综合征(OHSS)模型大鼠的疗效及作用机制。方法:20日龄Wistar大鼠60只,随机分成空白对照组、模型对照组、吲哚美辛组及zyyktc组,每组15只。除空白对照组外,其余3组采用改良的Ujioka方法建立OHSS模型,分别灌胃给予相应剂量的药物,于用药第7天处死大鼠,称取卵巢重量,肉眼、光镜观察卵巢组织形态学改变,采用放射免疫法检测大鼠血清E2水平,凝固法测定血浆纤维蛋白原(Fib)含量,ELISA法检测血浆组织型纤溶酶原激活物(t-PA)、纤溶酶原激活物抑制剂-1(PAI-1)及D-二聚体(D-D)含量。结果:zyyktc能显著降低OHSS大鼠血清E_2(P<0.01)含量和血浆D-D、Fib、tPA、PAI-1水平(均P<0.01),改善卵巢组织形态学变化。结论:zyyktc可有效调节OHSS大鼠凝血纤溶系统,改善血液循环,从而对OHSS发挥治疗作用。
Objective: To study the curative effect and mechanism of ziyktc on ovarian hyperstimulation syndrome (OHSS) model rats. Methods: Sixty Wistar rats aged 20 days were randomly divided into blank control group, model control group, indomethacin group and zyyktc group, 15 rats in each group. In addition to the blank control group, the other three groups using the improved Ujioka method to establish OHSS model, were given the corresponding dose of drug gavage, the rats were sacrificed on the 7th day, weighed ovarian weight, macroscopic, light microscopy ovarian tissue morphology The level of serum E2 was detected by radioimmunoassay, the content of fibrinogen (Fib) was measured by coagulation method. The levels of plasma tissue plasminogen activator (t-PA), plasminogen activator inhibitor Agent-1 (PAI-1) and D-dimer (DD) content. Results: zyyktc significantly reduced the level of E_2 (P <0.01) and the levels of D-D, Fib, tPA and PAI-1 in OHSS rats (all P <0.01), and improved ovarian tissue morphological changes. Conclusion: zyyktc can effectively regulate the coagulation and fibrinolytic system of OHSS rats, improve blood circulation, and thus play a therapeutic role in OHSS.
