目的了解包含WW域的氧化还原酶(WWOX)基因在儿童白血病中的表达,分析WWOX基因与儿童白血病的关系;探讨WWOX基因在儿童白血病中的发病机制。方法收集湖南省人民医院和长沙市其他医院2009年1月-2011年3月白血病患儿51例作为白血病组,同期非白血病患儿10例作为对照组,本研究经患儿监护人知情同意,采用巢式-聚合酶链反应检测2组儿童骨髓单个核细胞中WWOX mRNA表达,应用SPSS 17.0统计软件进行数据分析。结果白血病组患儿WWOX mRNA表达阳性率明显低于对照组(31.37%vs 100.00%,P<0.01);白血病缓解组患儿WWOX mRNA表达阳性率亦明显低于对照组(33.33%vs100.00%,P<0.01),2组比较差异有统计学意义;白血病组患儿WWOX mRNA表达阳性率在不同性别、年龄、白血病类型、治疗阶段、肿瘤负荷组间比较差异均无统计学意义(Pa>0.05)。结论儿童白血病患儿中WWOX mRNA的表达阳性率较低;WWOX mR-NA的表达阳性率可能与儿童白血病的发生发展有关。
OBJECTIVE: To investigate the expression of WWOX gene in children with leukemia, and to analyze the relationship between WWOX gene and childhood leukemia and to explore the pathogenesis of WWOX gene in childhood leukemia. Methods Hunan Provincial People’s Hospital and other hospitals in Changsha from January 2009 to March 2011 51 cases of leukemia in children as leukemia group, 10 cases of non-leukemia in the same period as a control group, the study informed consent of the guardian of children with The expression of WWOX mRNA in bone marrow mononuclear cells from two groups of children was detected by nested-polymerase chain reaction. The data were analyzed by SPSS 17.0 statistical software. Results The positive rate of WWOX mRNA expression in leukemia children was significantly lower than that in the control group (31.37% vs 100.00%, P <0.01). The positive rate of WWOX mRNA in leukemia children was significantly lower than that in the control group (33.33% vs 100.00% , P <0.01). There was significant difference between the two groups (P> 0.05). The positive rate of WWOX mRNA in leukemia group was not significantly different between different gender, age, leukemia type, treatment stage and tumor burden group (Pa> 0.05). Conclusions The positive rate of WWOX mRNA expression in children with childhood leukemia is low. The positive rate of WWOX mR-NA expression may be related to the occurrence and development of childhood leukemia.
