从龙眼中分离获得1个多聚半乳糖醛酸酶(PG)基因,命名为DlPG1。DlPG1基因与多种物种PG基因氨基酸序列有较高的同源性,其中与同属于无患子科的荔枝同源性最高,为95%,且与荔枝的亲缘关系最近。DlPG1基因包含PG基因特有4个保守区中的3个,DlPG1属于E进化支PG基因家族成员。环剥摘叶处理可以显著促进龙眼幼果脱落,对照累积落果率为6.12%,处理为89.85%,相对落果率高峰出现在处理后第5天。处理的离区DlPG1基因表达量始终高于对照,且表达量高峰出现在处理后第4天。基因表达量的高峰早于相对落果率高峰出现,据此推测,DlPG1基因可能是调控龙眼幼果脱落的关键基因。截至目前,DlPG1基因是首例在龙眼中发现的属于E进化支的可能与器官脱落相关的PG基因。
A polygalacturonase (PG) gene was isolated from longan and named DlPG1. The DlPG1 gene has high homology with the PG gene of many species. The homology of the DlPG1 gene is the highest with 95% for the lychee belonging to the same family, and the closest relationship with the litchi. The DlPG1 gene contains 3 of the 4 conserved regions unique to the PG gene, and DlPG1 belongs to the member of the E evolutionary PG gene family. Girdling leaves treatment can significantly promote the longan off, the control cumulative decapitation rate was 6.12%, deal with 89.85%, the relative deciduous peak appeared in the fifth day after treatment. The expression level of DlPG1 gene was always higher than that of the control, and the peak of expression appeared on the 4th day after the treatment. The peak of gene expression was earlier than the peak of relative fruit drop rate. It was speculated that DlPG1 gene might be the key gene regulating the shedding of young fruit of longan. Up to now, the DlPG1 gene is the first PG gene in the longan that belongs to the E clade and may be involved in exfoliation of the organ.
