【目的】准确检测口腔鳞状细胞癌及正常口腔粘膜中人乳头瘤病毒 16型 (HPV16)的感染状况 ,探讨人乳头瘤病毒 16型与人口腔鳞状细胞癌之间的关系。【方法】从 30例未经治疗的口腔鳞状细胞癌患者肿瘤组织及 30例健康志愿者正常口腔粘膜切取标本 ,液氮冷冻保存 ;常规提取组织DNA ,设计HPV16E7转化基因特异性引物 ,进行聚合酶链反应 ,对PCR结果进行检测 ,记录结果。【结果】 30例口腔鳞状细胞癌患者中检测到 11例HPV16阳性 ,感染率 36 7% ;30例正常对照组中检测到 4例HPV16阳性 ,感染率 11 1%。经 χ2 检验 ,两者差别有显著性。【结论】人乳头瘤病毒 16型与人口腔鳞状细胞癌有一定关系 ,但HPV16在人口腔鳞状细胞癌发生发展过程中所起的作用还有待进一步研究。
【Objective】 To accurately detect the infection status of human papillomavirus type 16 (HPV16) in oral squamous cell carcinoma and normal oral mucosa, and to explore the relationship between human papillomavirus 16 and human oral squamous cell carcinoma. [Methods] Specimens were harvested from normal oral mucosa of 30 untreated patients with oral squamous cell carcinoma and 30 healthy volunteers and cryopreserved in liquid nitrogen. Tissue DNA was routinely extracted and HPV16E7 specific primers were designed for polymerization. Enzyme chain reaction, detection of PCR results, and recording of results. 【Results】 Among the 30 patients with oral squamous cell carcinoma, 11 were positive for HPV16 and the infection rate was 367%. In the 30 normal controls, 4 were positive for HPV16 and the infection rate was 11%. After the χ2 test, the difference was significant. 【Conclusion】 Human papillomavirus 16 has a certain relationship with human oral squamous cell carcinoma, but the role of HPV16 in the development of human oral squamous cell carcinoma remains to be further studied.
