目的 探讨洛伐他汀对脂多糖(LPS)诱导的肾小球系膜细胞(MsC)表达炎性细胞因子的影响及其机制。方法 分别以RT-PCR和凝胶迁移率法(EMSA)的方法观察系膜细胞表达炎性细胞因子白细胞介素(IL)-1β、IL-6、肿瘤坏死因子(TNF)-α及单核细胞趋化蛋白(MCP)-1mRNA及核因子NF-κB的活化。结果 在洛伐他汀作用下4种细胞因子mRNA的表达均呈不同程度减弱,LPS诱导的MsC NF-κB活化也被抑制,而外加甲羟戊酸及法呢醇则可对抗这种抑制作用。结论 洛伐他汀可以通过抑制系膜细胞NF-κB的活性,降低炎性细胞因子的表达。为进一步认识和评价他汀类药物在肾脏疾病防治中的作用提供了理论及实验依据。
Objective To investigate the effect and mechanism of lovastatin on the expression of inflammatory cytokines in glomerular mesangial cells (MsC) induced by lipopolysaccharide (LPS). Methods The expressions of inflammatory cytokines IL-1β, IL-6, tumor necrosis factor (TNF) -α and mononuclear cells in mesangial cells were observed by RT-PCR and EMSA respectively. Activation of Cytokinin (MCP) -1 mRNA and NF-κB. Results The mRNA expressions of all four cytokines decreased to some extent under the action of lovastatin, and the activation of MsC NF-κB induced by LPS was also inhibited. However, the addition of mevalonate and farnesol could counteract this inhibitory effect. Conclusion Lovastatin can reduce the expression of inflammatory cytokines by inhibiting the activity of NF-κB in mesangial cells. This study provides a theoretical and experimental basis for further understanding and evaluating the role of statins in the prevention and treatment of renal diseases.
