目的探讨腺性膀胱炎及膀胱移行细胞癌中膜型基质金属蛋白酶1、5(MT1MMP、MT5MMP) mRNA及基质金属蛋白酶抑制剂2(TIMP2) mRNA的表达与膀胱肿瘤恶性程度和侵袭性之间的关系及其临床意义。方法采用逆转录聚合酶链式反应(RT-PCR)方法检测36例膀胱移行细胞癌(BTCC)和10例腺性膀胱炎及10例正常膀胱组织中MT1MMP、MT5MMP mRNA、TIMP2 mRNA的表达。结果MT1MMP mRNA、MT5MMP mRNA在BTCC中的表达明显高于腺性膀胱炎和正常膀胱组织中的表达(MT1MMP均值腺性膀胱炎0.661,膀胱肿瘤G21.569、G31.613,正常膀胱组织0.549,P<0.001;MT5MMP均值腺性膀胱炎0.574,膀胱肿瘤G21.508、G31.502,正常膀胱组织0.556,P<0.001),TIMP2 mRNA在BTCC中的表达则明显低于腺性膀胱炎及正常膀胱组织(均值膀胱肿瘤G21.391、G31.489,腺性膀胱炎1.797,正常膀胱组织1.958,P<0.001),腺性膀胱炎及正常膀胱组织中的表达间差异无统计学意义(P>0.05)。TIMP2 mRNA随肿瘤分级的增高而明显减少(P<0.001),但与分期无关(MT1MMP均值Tis~T11.428,T2~T41.628;MT5MMP均值Tis~T11.315,T2~T41.588;TIMP2均值Tis~T11.479,T2~T41.420;P>0.05)。结论MT1MMP mRNA、MT5MMP mRNA高表达与BTCC的恶性程度呈正相关,TIMP2 mRNA表达与BTCC的恶性程度呈负相关。MT1MMP mRNA、MT5MMP mRNA可能成为评估BTCC恶性程度及预后的指标。
Objective To investigate the relationship between the expression of membrane-type matrix metalloproteinase 1,5 (MT1MMP, MT5MMP) mRNA and matrix metalloproteinase 2 (TIMP2) mRNA in cystitis glandularis and bladder transitional cell carcinoma and the malignancy and invasiveness of bladder tumor Relationship and its clinical significance. Methods The expression of MT1MMP, MT5MMP mRNA and TIMP2 mRNA in 36 cases of transitional cell carcinoma of bladder (BTCC), 10 cases of cystitis glandula and 10 cases of normal bladder tissues were detected by reverse transcriptase-polymerase chain reaction (RT-PCR). Results The expressions of MT1 mRNA and MT5 mRNA in BTCC were significantly higher than those in cystitis glandula and normal bladder tissue (MT1MMP mean glandular cystitis 0.661, bladder tumor G21.569, G31.613, normal bladder tissue 0.549, P <0.001; MT5MMP mean glandular cystitis 0.574, bladder tumor G21.508, G31.502, normal bladder tissue 0.556, P <0.001). The expression of TIMP2 mRNA in BTCC was significantly lower than that in cystitis glandula and normal bladder tissue There was no significant difference between the two groups (P> 0.05), between cystitis glandularis and normal bladder tissues (mean bladder cancer G21.391, G31.489, cystitis glandularis 1.797, normal bladder tissue 1.958, P <0.001) . TIMP2 mRNA was significantly decreased with the increase of tumor grade (P <0.001), but not with staging (MT1MMP mean Tis ~ T11.428, T2 ~ T41.628; MT5MMP mean Tis ~ T11.315, T2 ~ T41.588; Mean Tis ~ T11.479, T2 ~ T41.420; P> 0.05). Conclusion The high expressions of MT1MMP mRNA and MT5MMP mRNA are positively correlated with the malignant degree of BTCC. The expression of TIMP2 mRNA is negatively correlated with the malignancy of BTCC. MT1MMP mRNA and MT5MMP mRNA may be used as indicators to evaluate the malignancy and prognosis of BTCC.
