目的:建立超高效液相色谱法测定酸水解前后化橘红中柚皮苷、野漆树苷、柚皮素、芹菜素的含量。方法:分别以超声法和酸水解法提取样品,超高效液相色谱法检测。采用WATERS ACQUITY UPLC色谱系统,ACQUITY BEH C18色谱柱(2.1mm×50mm,1.7μm),以甲醇-0.1%乙酸水溶液为流动相梯度洗脱,流速0.5mL/min,柱温30℃,检测波长283nm,289nm,337nm。结果:酸水解前可同时测定柚皮苷、野漆树苷、柚皮素、芹菜素的含量,酸水解后柚皮苷、野漆树苷分别转化为柚皮素、芹菜素,4种黄酮与其它成分均达到良好的分离。结论:该方法高效、快速、灵敏,为化橘红的抗氧化活性研究及其新制剂的开发提供参考。
OBJECTIVE: To establish a method for the determination of naringin, naringin, naringenin and apigenin before and after acid hydrolysis by ultra performance liquid chromatography. Methods: Samples were extracted by sonication and acid hydrolysis respectively and detected by ultra performance liquid chromatography. The mobile phase was eluted with a mobile phase of methanol-0.1% acetic acid at a flow rate of 0.5 mL / min on a ACQUITY BEH C18 column (2.1 mm × 50 mm, 1.7 μm) using a WATERS ACQUITY UPLC system. The detection wavelength was 283 nm , 289 nm, 337 nm. Results: The contents of naringin, naringin, naringenin and apigenin were determined simultaneously before acid hydrolysis. After acid hydrolysis, naringin and naringin were respectively converted to naringenin, apigenin, four flavones and others The ingredients are well separated. Conclusion: The method is efficient, rapid and sensitive. It provides a reference for the research on antioxidant activity of orange juice and the development of new preparation.
