目的:研究碱性成纤维生长因子(bFGF)、整合素αⅤβ3和核因子κB(NFκB)在人卵巢上皮性癌(OEC)组织中的表达及其与血管生成的关系·方法:原发性人卵巢上皮癌(hOEC)36份、卵巢良性肿瘤及正常卵巢15份,应用免疫组化方法(ABC法)检测bFGF、整合素(integrin)αⅤβ3、NFκB、微血管密度(microvesseldensity,MVD)在OEC、卵巢良性肿瘤及正常卵巢组织中的表达及其与临床病理特征的关系.结果:bFGF、αⅤβ3、NFκB和MVD在各种组织中的表达,36份OEC中的阳性表达率分别为66·67%、66·67%、72·22%和(43·98±33·73),在良性肿瘤组织中的表达率分别20·00%、6·67%、26·67%和(13·08±9·07);在正常卵巢组织的表达率分别为0·00%、0·00%、13·33%和(7·10±5·42),OEC组各指标的阳性率均显著高于良性卵巢肿瘤组及正常对照组(P<0·05).OEC中bFGF、αⅤβ3、NFκB、MVD随着肿瘤分化程度的降低、FIGO分期的增加、肿瘤转移范围扩大,各指标的表达均逐步增高(P<0·05).OEC中bFGF、αⅤβ3、NFκB与MVD的表达的相关系数分别为0·69、0·67、0·62、各指标与MVD均显著相关(P<0·01).NFκB阳性表达者bFGF、αⅤβ3的阳性表达率分别为92·86%和85·71%,显著高于NFκB阴性表达者bFGF、αⅤβ325%的表达率(P<0·05).结论:bFGF,αⅤβ3,NFκB在OEC中的表达显著增高,与OEC血管生成密切相关.bFGF、αⅤβ3在OEC中的表达与NFκB表达呈正相关,NFκB可能通过对bFGF、αⅤβ3的正向调节作用,进而影响OEC血管生成.
Objective: To investigate the expression of basic fibroblast growth factor (bFGF), integrin αVβ3 and NFκB in human ovarian epithelial carcinoma (OEC) and its relationship with angiogenesis · Methods: Primary human 36 cases of ovarian epithelial carcinoma (hOEC), 15 cases of ovarian benign tumor and 15 cases of normal ovary. The expressions of bFGF, integrin αVβ3, NFκB and microvessel density (MVD) in OEC and ovary were detected by immunohistochemistry (ABC method) The correlation between the expression of bFGF, αVβ3, NFκB and MVD in various tissues and the positive rates of the expression in 36 OECs were 66.67% 66.67%, 72.22% and 43.98 ± 33.73, respectively. The expression rates in benign tumor tissues were 20.00%, 67.6%, 26.67% and 13.08 ± 9% respectively · 07). The positive rates of OEC were significantly higher than those of benign ones in normal ovarian tissues (0 · 00%, 0 · 00%, 13 · 33% and (7 · 10 ± 5 · 42) Ovarian tumor group and normal control group (P0.05) .The bFGF, αⅤβ3, NFκB and MVD in OEC decreased with the degree of tumor differentiation, the increase of FIGO stage, the expansion of tumor metastasis (P0.05) .The correlation coefficients of bFGF, αVβ3, NFκB and MVD in OEC were 0.69,0.67 and 0.62 respectively, and all the indexes and MVD were significant (P <0.01) .The positive rates of bFGF and αVβ3 in positive NFκB expression were 92.86% and 85.71%, respectively, which were significantly higher than those in bFGF and αVβ325% · 05) .Conclusion: The expressions of bFGF, αVβ3 and NFκB were significantly increased in OEC and closely related to OEC angiogenesis.The expressions of bFGF and αVβ3 in OEC were positively correlated with the expression of NFκB, which may be related to the positive expression of bFGF and αVβ3 Regulatory effect, thereby affecting OEC angiogenesis.
