目的观察白芍总苷(TGP)对非酒精性脂肪性肝病(NAFLD)大鼠及脂肪因子apelin和visfatin的影响。方法以果糖-高脂诱导非酒精性脂肪性肝病大鼠模型,模型大鼠随机分为模型组,二甲双胍组(200 mg.kg-)1,TGP高、低剂量组(200,100 mg.kg-)1,另设正常对照组。用药4周后观察各组大鼠空腹血糖(FBG)、胰岛素(Fins)、胰岛素敏感指数(ISI)、胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、甘油三酯(TG)、游离脂肪酸(FFA)、apelin-36、visfatin、天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、谷胱甘肽转移酶(GST)和肝脏指数。结果 TGP高、低剂量组FBG、Fins、LDL-C、TG、apelin-36和visfatin均明显降低,ISI和HDL-C含量明显升高(P<0.01或P<0.05),TGP高剂量组能显著降低ALT、AST和GST的活性及肝脏指数(P<0.01),TGP低剂量也能显著降低A ST和GST的活性(P<0.01)及肝脏指数(P<0.05)。结论 TGP能改善果糖-高脂诱导NAFLD大鼠糖脂代谢异常及拮抗胰岛素抵抗,增强胰岛素敏感性,下调apelin和visfatin的表达,改善肝功能。
Objective To observe the effects of total glucosides of paeony (TGP) on non-alcoholic fatty liver disease (NAFLD) rats and adipokines apelin and visfatin. Methods The model rats were randomly divided into model group, metformin group (200 mg.kg -) 1, high and low TGP group (200, 100 mg.kg -), high fructose - induced fatty liver disease 1, another set of normal control group. Four weeks after treatment, fasting blood glucose (FBG), insulin (Fins), ISI, TC, LDL-C Triglyceride (TG), free fatty acid (FFA), apelin-36, visfatin, aspartate aminotransferase (AST), alanine aminotransferase (ALT), glutathione transferase (GST) and liver index. Results The levels of FBG, Fins, LDL-C, TG, apelin-36 and visfatin in high and low TGP groups were significantly decreased, while the levels of ISI and HDL-C were significantly increased (P <0.01 or P < The activity of ALT, AST and GST and the index of liver (P <0.01) were significantly decreased. Low doses of TGP could significantly reduce the activity of AST and GST (P <0.01) and the index of liver (P <0.05). Conclusion TGP can improve glucose and lipid metabolism, antagonize insulin resistance, enhance insulin sensitivity, decrease apelin and visfatin expression and improve liver function in fructose-high fat-induced NAFLD rats.
