目的:研究GGTase-1在人脑胶质瘤中的表达,并分析其表达与胶质瘤病理级别的关系,探讨GG-Tase-1对人脑胶质瘤发生、发展所起的作用,从而为胶质瘤的治疗提供一定的理论依据。方法:分别应用半定量RT-PCR法和WesternBlot法检测15例人脑胶质瘤标本和4例正常脑组织中的GGTase-1β亚单位(GGTβ)mR-NA水平和蛋白水平;应用免疫组织化学法检测29例人脑胶质瘤组织标本和6例正常脑组织中GGTβ蛋白表达与定位情况。结果:GGTβmRNA和蛋白在各级人脑胶质瘤和正常脑组织均可表达,但在人脑胶质瘤中的表达明显高于正常脑组织(P<0.05);并且GGTβmRNA和蛋白在人脑胶质瘤Ⅲ~Ⅳ级组中的表达明显高于Ⅰ~Ⅱ级组(P<0.05)。结论:①GGTase-1在人脑胶质瘤和正常脑组织中均表达,但在人脑胶质瘤中呈明显高表达,可能与人脑胶质瘤的发生、发展有关。②GGTase-1的表达与人脑胶质瘤的病理分级相关。
OBJECTIVE: To study the expression of GGTase-1 in human glioma and to analyze the relationship between the expression of GGTase-1 and the pathological grade of glioma and to explore the role of GG-Tase-1 in the occurrence and development of human glioma. To provide some theoretical basis for the treatment of glioma. Methods: The mRNA and protein levels of GGTase-1β subunit (GGTβ) mR-NA in 15 human glioma specimens and 4 normal brain tissues were detected by semi-quantitative RT-PCR and Western Blot respectively. Immunohistochemistry Method to detect the expression and localization of GGTβ protein in 29 human glioma tissue samples and 6 normal brain tissues. Results: GGTβmRNA and protein were expressed in all kinds of human gliomas and normal brain tissues, but were significantly higher in human gliomas than in normal brain tissues (P <0.05); and GGTβmRNA and protein in human brain The expression of glioma in grade Ⅲ ~ Ⅳ group was significantly higher than in grade Ⅰ ~ Ⅱ group (P <0.05). Conclusions: ①GGTase-1 is expressed in human glioma and normal brain tissue, but it is highly expressed in human glioma, which may be related to the occurrence and development of human glioma. The expression of GGTase-1 was correlated with the pathological grade of human glioma.
