目的 观察水杉总黄酮 (FMG)对大鼠血小板聚集活性、血小板 5 HT释放反应、血浆NO含量及血液流变性的影响。方法 比浊法测定大鼠血小板聚集活性、荧光光度法测定血小板 5 HT释放反应、分光光度法测定血浆NO含量、血液流变全自动分析仪检测血液流变性。结果 FMG抑制大鼠血小板聚集活性 ,抑制血小板 5 HT释放 ,升高血浆NO含量 ,降低急性血淤大鼠的全血比粘度、血浆比粘度、红细胞电泳时间、红细胞压积、红细胞计数、细胞方程K值 ,改善大鼠的血液流变性。结论 FMG有抗血小板聚集活性、改善血液流变性的作用。其抗血小板聚集的作用机制可能与抑制血小板释放反应、增加体内NO合成及Ca2 + 拮抗作用有关
Objective To observe the effect of total flavones (FMG) on platelet aggregation activity, platelet 5 HT release, plasma NO content and hemorheology in rats. Methods Turbidimetric method was used to determine the platelet aggregation activity in rat platelets. The platelet 5 HT release reaction was measured by spectrofluorimetry. The content of NO in plasma was measured by spectrophotometry. The hemorheology was detected by automatic analyzer. Results FMG inhibited platelet aggregation, inhibited platelet 5 HT release, increased plasma NO content, decreased whole blood viscosity, plasma specific viscosity, erythrocyte electrophoresis time, hematocrit, erythrocyte count, cell equation K value, improve the rat’s blood rheology. Conclusion FMG has anti-platelet aggregation activity and improves the hemorrheology. Its anti-platelet aggregation mechanism may be related to inhibition of platelet release reaction, increased NO synthesis and Ca2 + antagonism
