目的:研究猪型布鲁氏菌外膜蛋白基因omp19的序列性质,同时,进行目的基因的原核表达。方法:以猪型布鲁氏菌基因组作为模板进行基因序列的扩增,利用大肠杆菌原核表达系统进行重组表达。结果:猪型布鲁氏菌外膜蛋白基因omp19的开放阅读框序列全长为534 bp,演绎177个氨基酸。诱导表达的重组蛋白质的理论分子量为17.6 kD,而且实际大小与理论值一致。结论:成功表达了Omp19分子,为进一步研究Omp19分子的免疫保护作用奠定了基础。
OBJECTIVE: To study the sequence properties of the outer membrane protein gene omp19 of Brucella swine and to perform the prokaryotic expression of the target gene. Methods: The genomic sequence of Brucella swine was used as a template to amplify the gene sequence. The recombinant plasmid was expressed in E. coli prokaryotic expression system. Results: The open reading frame of omp19 gene of Brucella abortus was 534 bp, which deduced 177 amino acids. The theoretical molecular weight of the induced recombinant protein was 17.6 kD, and the actual size was consistent with the theoretical value. Conclusion: The Omp19 molecule was successfully expressed, which laid the foundation for the further study of the immunoprotection of Omp19.
